Abstract
PURPOSE: To search for new alternatives for in-vitro corneal epithelial cell growth.
METHODS: Using PDLLA (poly D L Lactic Acid) polymer and adding cationic polymer Eudragit RL 100 in different ratios, microparticles were prepared and studied for their morphology using optical microscopy and scanning electron microscopy (SEM). Biodegradable scaffold membranes were prepared at room temperature, which were characterized for their surface morphology, surface charge analysis, porosity, water retention capacity and cell growth profile. Suitability of the modified scaffolds in promoting cell growth and attachment were assessed by cell growth and proliferation studies. Corneal epithelial cell lines from rabbit(SIRC cell lines)were used. Extraction of primary corneal epithelial cells from laboratory mice cornea was also attempted.
RESULTS: The biodegradable scaffolds prepared from PDLLA-Eudragit polymers in the ratio of 70:30 were found to be suitable for the corneal epithelial cell culture.
